Adaptive design to find the best diagnostic biomarker.

One objective in diagnostic study is to find the best biomarker among several competing biomarkers and then to show superiority of this biomarker to the standard one. For ethical and economical reasons, a adaptive two stage design will be proposed. At first stage we will estimate the best biomaker by areas under the receiver operating characteristic (ROC) curves(AUC), and stop the study if there is enough evidence for accepting or rejecting superiority. Otherwise, the second stage is carried out with the best biomarker estimate and the standard biomarker. Sample Size of second stage will be estimated based information from the first stage. The performance of the design will be investigated numerically.
